STANDARD ANAEROBIC CULTURE BOTTLE



The biochemical receptor is mainly composed of liquid silicone rubber and xylenol blue; the liquid culture medium is mainly composed of peptone (5g/L), beef extract powder (3g/L), hemin (0.005g/L), yeast extract powder (3g/L), and sodium polyisocyanate (0.15g/L); the gas in the culture bottle is sterile air (oxygen content is about 21%, nitrogen content is about 78%).

RESIN AEROBIC CULTURE BOTTLE

The biochemical receptor is mainly composed of liquid silicone rubber and xylenol blue; the liquid culture medium is mainly composed of peptone (5g/L), beef extract powder (3g/L), hemin (0.005g/L), yeast extract powder (3g/L), sodium polyanethol sulfonate (0.15g/L), and macroporous adsorption resin (3.5g/bottle); the gas in the culture bottle is sterile air (oxygen The gas content is about 21% and the nitrogen content is about 78%).

LIMITATIONS OF THE TEST METHOD

1. This product is not suitable for the cultivation of anaerobic bacteria and mycobacteria.
2. If there are microbial inhibitors in the sample, there is a possibility of missed detection.
3. Some rare fastidious microorganisms may be difficult to grow in culture bottles. If there is any doubt, other cultural methods should be used.
4. Pneumococcus is prone to autolysis if not transferred immediately after being reported positive.
5. A rare phenomenon may occur where bacteria grow in the culture bottle but fail to report a positive result because the concentration of CO2 produced is not high enough.
6. When using resin aerobic culture bottles, if the sample contains antibiotics, bacteria may occasionally grow in the culture bottle but fail to report positive. In this case, the culture medium can be used for subculture.
7. If the sample inoculated in the culture bottle is insufficient, some strains that may be sensitive to anticoagulants (sodium polyanethol sulfonate), such as Haemophilus influenzae, Neisseria meningitidis, and gonorrhea strains, may not grow or produce low concentrations of CO2.

USAGE METHOD OF BLOOD CULTURE COLLECTION

- Sterile blood sampling (i.e. hand hygiene, sterile dressing pack, sterile gloves, and clean skin with chlorhexidine)

- Take from fresh venepuncture or alternatively newly inserted peripheral cannulae and central lines

- If line sepsis is suspected cultures should be taken from the central line and peripherally
 

USAGE METHOD OF BLOOD CULTURE COLLECTION

- Blood placed into aerobic and anaerobic culture medium
remove the dust cap from culture bottles and top swabbed with alcohol (and allowed to dry) before blood is injected

- Maximum yield if the tube is neither under nor over-filled (e.g. 9 mL in adult tubes)

- Fill blood culture tubes before other blood tubes

OTHER INFORMATION NUMBER OF CULTURES

- Two sets of cultures from separate sites before starting antibiotics are ideal

- 3 cultures have a 96% sensitivity in detecting bacteremia
more than 4 cultures generally offer little additional benefit
additional cultures may be beneficial if suspected endocarditis or pre-existing antimicrobial use

- Antibiotics should not be withheld pending culture in critically ill septic patients

INCUBATION AND CULTURE

- Incubation at room temperature initially to avoid killing temperature-sensitive bacteria

- Turbid appearance may indicate microbial growth - bottles are placed into holders with light passed through - an alarm sounds when significant microbial growth interrupts the passage of light through bottles

- All samples are subcultured onto agar plates to test purity, allow organism identification, and for antibiotic sensitivity testing

CONTAMINATION

suspect if single, random cultures of Bacillus species, coagulase-negative staphylococci or diphtheroid
less likely if grown in more than one bottle (another reason for taking multiple cultures from separate sites)
